PHARMACOLOGY:

Pharmacodynamics Nimesulide is a non-steroidal anti-inflammatory drug that selectively inhibits cyclooxygenase-2 but it appears to exert its therapeutic effects through variety of other mechanisms as well like inhibition of platelet activating factor (PAF), tumor necrosis factor- alpha (TNF-a), protease & histamine etc.

Pharmacokinetics

Oral absorption of Nimesulide is almost complete. Plasma binding is high (approximately 99%). Nimesulide is extensively metabolized to several metabolites which are excreted mainly in the urine (about 70%) and in feces (about 20%)

Nims tablet indication

Nimesulide, being an NSAID, exhibits anti-inflammatory & analgesic properties. It is indicated as second choice for the treatment of acute pain and primary dysmenorrhea.

SPECIAL WARNINGS AND PRECAUTIONS FOR USE:

Hepatic Effects: Rarely Nimesulide has been reported to be associated with serious hepatic reactions, including very rare fatal cases.  should have treatment discontinued. These patients should not be rechallenged wiut Nimesulide.

CONTRAINDICATIONS:

The use of Nimesulide is contraindicated in patients with hepatic impairment, children, active peptic ulcer disease, known hypersensitivity to the active substance, acetylsalicylic acid, or other prostaglandin synthetase inhibiting drugs.

ADVERSE REACTIONS:

Undesirable effects include gastrointestinal disorders, gastrointestinal bleeding, duodenal ulcers and perforation, gastric ulcer and perforation.

SIDE-EFFECTS:

At therapeutic doses Nimesulide is usually well tolerated. Occasional side effects include heartburn, nausea and epigastric pain. Serum rashes of allergic may rarely occur and bleeding from stomach, stomach ulcer.

Nims tablet DOSAGE AND ADMINISTRATION.

Usual oral dose in adults is one tablet twice daily for not more than 15 days OR As directed by the physician
